Clinical and Experimental Studies on Ginseng-Peony


Ginseng-Peony tablet is composed of herbs of treating cardiovascular disorders, including Radix Paeoniae Alba and saponins extracted from leaf and stem of ginseng.
The main indications of the tablet are coronary heart disease and angina pectoris. The clinical and experimental studies on the tablet are summarized as the follows.

Experimental Studies
Animal experiments indicate the following efficacy.
1). Coronary blood flow of isolated heart of guinea pig was increased by the tablet, with decreased oxygen consumption and release of coronary artery spasm. The vessel dilatation effect cou1d benefit the ischemic heart, the restoration of heart function and the perfusion of blood to the whole body.
2). The acute ischemia of rat heart induced by pituitrin was modified by administration of “Shen Shao Tablet" The spasm of coronary artery was inhibited with increase in coronary blood flow and improvement of myocardium metabolism.
3). The survival time of mouse with ischemic myocardium, induced by hypoxia under normal pressure and injection of iso-protereno, was significantly prolonged by the tablet. The swimming time of mouse was also prolonged by the tablet. Both phenomena indicate the effects of anti-fatigue and anti—hypoxia.

Clinical Studies
1). Inhibition of platelet aggregation: The investigation on platelet activity and aggregation by electronic microscope revealed that platelet aggregates were significantly reduced by treatment. It is a clear indication of beneficial effect on antithrombosis.
2). Improvement of microcirculation.
3). Improvement in hemorrheological parameters, including serum relative viscosity and blood viscosity.
4). Increase in left ventricular output.

Safety Evaluation
1). Acute toxicity: The acute LD50 of mouse could not be reached by administration of maximum dose of the tablet. No death was found within seven days when mice were fed with a dose of 480 fold of human daily dose for 3 days.
2). Chronic toxicity: Fourteen health dogs were fed the tablet for 18 Weeks. The daily dosage was estab1ished as 10 and 20 fold of human adult dose.
Functions of liver, kidney and heart, erythrocytes count, leukocyte count, platelet count, hemoglobin level, leukocyte classification, were all monitored monthly. No abnormal signs were found. The dogs were healthy, active, and normal in appetite, urine and feces. The body weight was gained after the chronic to toxicity test. 
Pathological examination revealed that no gangrenous changes were found in heart, liver, spleen, kidney and stomach for both dose groups. No significant differences could be determined between the dose groups and the control. The only exceptional was a moderate fatty liver (No. 14 dog, control group). This is considered as the original problem of the dog.

Summary
The “Shen Shao Tablet"is considered as a drug of activating blood circulation to remove blood stasis. As experiments demonstrated, improvement of blood circulation and anti-hypoxia are the underline mechanism of action of the tablet. This is the basis for understanding of the clinical efficacy of the tablet in treatment for angiocardiopathy. The safety of the tablet is ensured by the above mentioned acute and chronic toxicity tests.
