Experimental Research on the Effect of Acupuncture on the EEG of Rat with


Objective: To study the mechanism of effect of acupuncture on the rat with epilepsy.

Method: (l) 70 rats were randomly divided into two groups with l0 fats in the normal group and 60 rats in another group for electric stimulation; (2) Two needles were respectively inserted into the eye ground on both sides along the medial aspects of the eye ball as the electrodes; (3) The handle of both needles were connected with YSD-4 Multiple-Use Apparatus for producing epilepsy model. The stimulation parameters were:
  (1) frequency of 8Hz, wave duration of 4ms; (2) continuous waves, intensity of l0V stimulation duration of l0 minutes. After electrica1 stimulation, all of the rats showed an attack of rigid convulsion all over the body. Stimulation was applied once every 24 hours with l0 days in succession; (4) methods of Electroencephalography: locations of the electrodes were set by reference to the l0/20 System Standard of Intemational EEG Society and recording electrodes were respectively on C3 and C4; A1 and Al on both ears were the reference electrodes, CZ is linked with the ground; (5) 30 rats with attack of epileptic convulsion and typical epileptic electric discharge of cerebral electricity were randomly divided into model group, acupuncture group, and drug control group; (6) Method of treatment: Baihui (GV20), Dachui (GV14), Jinsuo (GV8), Yaoqi (EXB9), Jiuwei (CVl5), Juque (CVl4), Xiawan (CVl l0), Guanyuan (CV4), Hegu (Ll4), Jianshi (PC6), Fenglong (ST40) and Taichong (LR3) were selected. The needles were retained for 20 minutes and manipulated once again every 5 minutes. After l0 days of acupuncture treatment, electroencepha1ogram (EEG) was recorded.

Results and Conclusion: (l) Effect of acupuncture on the cerebra1 electricity of rats with epilepsy: compared with the normal group, the basic frequency of EEG decreased remarkably (P<0.0); the basic amplitude of EEG increased obviously (P<0.0l), and there appeared regular epileptic discharge with the highest amp1itude of wave being l70-340mv and higher electric discharge frequency being 7 - l4times/min in epi1epsy model group. Compared with the model group, acupuncture cou1d distinctively increase the basic frequency of cerebra1 e1ectrical activity in rats with epi1epsy (P<0.01) and could distinctively lower the average amplitude and highest amplitude of EEG (P<0.01); and distinctively reduce the discharge frequency (P<0.0l). The acupuncture group had significant differences in comparison with the drug group in improving the basic EEG frequency (P<0.05); also in decreasing the basic amp1itude, and average amplitude as well as highest amplitude of epileptic discharge (P<0.0l); (2) Effect of acupuncture on the waveform of EEG: acupuncture could remarkably raise the basic frequency of EEG and lower the average amp1itude and highest waves in rats with epilepsy. Meanwhile, it could a1so improve the waveform of EEG in rats with epilepsy.
